Flockscreen, Mycoplasma synoviae Stained Antigen Kits

x-OvO Limited is pleased to announce the launch of the FLOCKSCREEN Mycoplasma synoviae (MS) Slide Agglutination Test Kit.
 
The use of slide agglutination testing to demonstrate an immunological response to mycoplasma spp infections is one of the cornerstones of global poultry health control programmes.  Consistency of results together with an ability to interpret those results relative to well defined positive and negative controls is one of the critical factors that influence the diagnostic power of the test.
 
Each kit contains a 10 ml (400 tests) presentation of FLOCKSCREEN Mycoplasma synoviae stained antigen together with individual vials of 0.1ml lyophylised positive and negative control sera.  The control sera were developed exclusively for x-OvO Limited by IZS Venezie, Legnaro, Italy.  x-OvO Limited has applied proprietary staining technologies to purified antigen bulks to produce a class leading product with extremely low levels of autoagglutination, a common problem in commercial MS stained antigen preparations.  For the first time, veterinary diagnostic laboratories can obtain control sera developed specifically and certified for use with each individual kit, thus facilitating more accurate test interpretation and results comparison within and between individual kits.
 

The MS Slide Agglutination Test Kit compliments the MG Slide Agglutination Test Kit, FLOCKSCREEN MG, MS and MG/MS combined ELISA tests and a centralised Mycoplasma PCR diagnostic to provide poultry diagnostic laboratories and producers with the complete Mycoplasma diagnostic programme.

FLOCKSCREEN ART Differentiation ELISA Kit

Of the 4 defined subtypes, A and B subtypes of avian rhinotracheitis virus are the most common globally.  Protection against infection is achieved by the use both live and inactivated vaccines.  Such products are protective against homologous and heterologous infection even though they are monovalent in nature.  From a clinical management perspective, the diagnosis of the presence of a particular subtype of virus in the field and the application of a heterologous subtype vaccine will facilitate the best control strategy should differentiation of the bird’s immune response to the two antigens be possible.
 
Until now, ELISA diagnostic kits have measured the combined A and B subgroup responses to ART infection.  The only means to detect the presence of a particular subtype was by the use of PCR based diagnostics on tracheal swab samples.  Successful results are rare due to the extremely short time window where live infectious ART virus is present at the site of sampling (the trachea).
 
As an important aid to the most appropriate management of ART infections and vaccination, x-OvO Limited is delighted to offer the FLOCKSCREEN ART Differentiation ELISA Kit.  The kit features highly purified antigens and sera for both A and B subtypes, selected specially for their subtype specific responses, custom manufactured by IZS Venezie, Legnaro, Italy (National, OIE and FAO reference laboratory).  ELISA plates provided in the kit are coated with either A or B subgroup antigen, with a single sample subject to a double analysis.  Comparison of both results yields important information on the major immune response of the bird and hence the possibility of a particular subtype infection.  Once an appropriate diagnosis is made of the subtype of field infection present, the differential response to a heterologous vaccine can be monitored to ensure appropriate vaccine application.  This should lead to decreasing titres to the field virus as the level of field infection is controlled.
 
The kit is supplied in a 2 plate or 4 plate format, each plate capable of analysing 96 samples for signs of either A or B subgroup infection.

Infectious Salmon Anaemia Diagnostic Service

In response to the recent global outbreaks of infectious salmon anaemia virus (ISAV) in farmed salmon,
x-OvO Limited is pleased to offer a centralised diagnostic PCR and HE gene sequencing service to allow clients to identify and track the spread of ISAV.
 
The specific provision of elongated sequences (1200 nucleotides) from the variable region of the HE gene assists veterinarians, farmers and fish production companies in the identification of the potential source of their ISAV infection via statistical comparison of isolates from infected farms to possible infectious sources (e.g. the trout reservoir or genetic source stock).
 
The analysis is carried out at the IZS Venezie in Legnaro, Italy on both frozen tissue samples or those submitted in viral transport media.

Avian Adenovirus Genotyping Diagnostic Service

 
x-OvO Limited has developed a centralised genotyping diagnostic service to facilitate the identification of avian adenovirus from tissue samples taken from commercial poultry flocks.  The test will allow the determination of the degree of association between two isolates and thus provide statistically significant evidence of the likelihood of transmission (or not) of any such virus from a source flock to a progeny flock.  Association with genotypes know previously to cause significant pathologies is also possible.
 
x-OvO Limited is pleased to announce the launch of a novel genetic typing analysis service for avian adenovirus infections in domestic poultry.

Although often implicated in sub-optimal performance of breeding birds, clear diagnosis of the presence of pathogenic adenoviruses (and hence their appropriate control) has been difficult historically due to a reliance on serological typing methods. In a similar vein, determination of the relationship (or lack of) between source flock adenoviruses and those isolated in progeny flocks and the resultant causal relationship between any observed pathologies has been extremely problematic.
 
To facilitate a clarification of these possible relationships, x-OvO Limited has developed a centralised diagnostic service able to screen and classify different types of Fowl Adenovirus A based on hexon loop (L1) genotyping.
 
The hexon protein is the major capsid protein of the non-enveloped virion on which type, group and subgroup specific determinants are located. The hexon consists of conserved regions (pedestals), which are located inside the virion, and variable loops which protrude from the surface containing the type-specific neutralizing epitopes.  Due to this interaction with the immune system, the sequence identity between loop regions of different serotypes is low.
 
Using these properties of the virus, it is possible to group together different Fowl Adenovirus strains in 5 (A-E) Genogroups based on their hexon L1 sequences. Inside each Genogroup it is possible to distinguish different clades that may be associated with individual serotypes.  Pyrosequencing analysis of key variable regions allows the rapid determination of lack of association of any two analysed samples.  Should samples display characteristics that indicate possible association with each other, the use of boot strapping analysis on extended genetic sequences allows the investigator to arrive at statistically significant conclusions regarding the genotype of a strain and its association with another strain. Thus the technology is ideally suited to determine if an adenovirus isolated from one flock is genetically the same (and hence a candidate for vertical transmission) as a virus isolated from its source supply flock.
